You will need To perform the grain inoculations, you require a sterile environment. Air is full of impurities and so it is important to reduce the level of containments where possible. Using thin lengths of wood, nail these together to make four wall panels.

Cover these panels with plastic sheeting use staples to attach. Screw the four panels together using sealant along the joins. Run sealant between the base of the walls and the floor. Finally, tape a plastic sheet over the top of the room to form its ceiling.

The room should be able to be easily wiped down with a bleach solution. The room also requires a work bench and storage shelves for incubating your jars of spawn.

Mushroom Spawn

Most mushroom clean rooms have a HEPA filter installed to provide clean oxygenated air. This instructable uses a low tech approach and will give you alternative methods for minimising air contaminants.

Preserving jars are ideal for preparing mushroom spawn. If you are unable to buy suitable jars, then recycling old jars is even better we used 1 litre olive jars. Make sure you clean your jars thoroughly, inside and out. Taking a drill, make an 8mm hole in each jar lid. Later, this will allow the mushroom spawn to breath. Soak the grain for 24 hours in a bowl of water use enough water to cover the grain completely. If you are using larger grains, like wheat or corn, I found simmering the grain for 30 minutes instead of soaking helped to prevent growth of unwanted moulds.

Rinse and drain the grain thoroughly. If required see Step 8 Liquid Inoculation Methodsprepare a jar of clean water to be sterilised with a few coins added - these help to cut up grain spawn later when shaking the jarfit lid and cap with aluminium foil. Add approximately 3 litres of water, pouring it carefully around the jars. Well, sometimes but it's not ideal. A certain amount of spawn can inoculate a much greater amount of substrate; resulting in many more mushrooms than if you used spawn alone. The rest of this article explains some of the ins and outs of mushroom spawn.

First we'll take a look at types of spawnand then move on to which type of spawn you should use. We'll end with how to acquire and store it. Any material with mycelial growth used to propagate mushrooms is considered 'spawn'. However, you'll usually find that spawn comes in one of these forms:. Sawdust Spawn is sterilized sawdust that has been inoculated with mycelium, often by grain spawn. The sawdust is usually made out of some type of hardwood with pieces neither too large nor too fine a few mm in diameter.

Sawdust spawn can be used to inoculate logs, outdoor mushroom beds, pasteurized straw, cardboard, and a variety of other substrates. It's also used to inoculate wooden dowels to create plug spawn. One of the main advantages of mushroom spawn in the form of sawdust is the size of the particles.

Because they are so small and numerous, there are many more inoculation points for the mycelium to grow into the substrate. This results in faster mycelium colonization with less time for contaminants to take hold. A disadvantage of sawdust spawn is that, on its own, its not as nutritious for mushroom growing as one would like. Thus you may be disappointed with your yield if you try to grow mushrooms from straight sawdust.

Mushroom growing kits made from sawdust are usually enriched with bran or some other source of nitrogen to increase yields.

Grain spawn is sterilized grain that has been inoculated with spores or a sterile culture of mycelium. Many types of grain can be used with rye and millet being some of the most common. Other choices are corn, wheat, and different cereal grains. I've even had good luck with popcorn! Grain spawn can be used to create sawdust spawn, more grain spawn, or inoculate all sorts of pasteurized substrates such as straw.Gourmet mushrooms such as oyster or shiitake mushrooms can be expensive to buy, but they are easy to grow.

Most people use commercially available mushroom spawn for the kind of mushroom they wish to grow. They mix the spawn into a growing medium such as wood dust or sawdust and harvest the mushrooms a few weeks later. Going back a step and making the mushroom spawn is more difficult. A good source of spores and laboratory conditions for a sterile environment are needed.

Once these initial conditions are in place, mushroom spawn can be made in large quantities and with excellent results.

Mushroom spawn seeds for growing mushrooms can be made under sterile conditions with grain seeds as a substrate. Mushroom spores, obtained from the inside of a mushroom cap, are initially grown in small dishes on gelatin.

The dishes and the growth media have to be sterilized and kept in a sterile environment. When the spores have produced mycelial growth, pieces of this culture are placed in sterilized jars with boiled grain seeds such as millet, rye or wheat.

The fungal mushroom culture penetrates the seeds completely after a few weeks. The seeds can then be used as mushroom spawn and mixed in with sawdust or other organic material to produce mushrooms.

Mushrooms are the fruits of the mycelium fungus, and the spores produced by the mushrooms are a kind of seed.

Scattered spores often don't produce any fungus because they need ideal conditions to grow. To make mushroom spawn, the ideal conditions required by the spores have to be created in a controlled setting. Once the spores have generated a well-established growth of fungus, the fungus culture can be transferred to grains. Placing mushroom spores onto a sterile medium to start the fungal growth and then transferring the culture to millet seeds is a good way to make oyster mushroom spawn.

Gelatin with a small amount of sugar, boiled for sterilization and poured into small, sterile jars makes a good starting medium.

Spores from the inside of the cap of an oyster mushroom placed on the gelatin with sterile tweezers will produce mycelium growth within about a week. Once the mycelium culture is established in the jars, the culture can be cut into pieces with a sharp, sterile knife. Millet seeds have to be soaked overnight and then boiled for an hour to sterilize them. After they are cool, they are placed in sterile mason jars together with the pieces of the mycelium culture, and the jars are well-shaken to mix the seeds and the culture.

Jars should be kept in the dark at room temperature for 10 to 20 days until the mycelium has completely penetrated the millet seeds. These seeds are the mushroom spawn seeds used for growing mushrooms in organic material. The gelatin medium and the millet grains are very fertile environments for the growth of all kinds of fungi and bacteria.

Contaminating spores and organisms from the surroundings in the home can easily come into contact with the spawn production materials.

Mushroom spores will only grow in the absence of competing organisms so the maintenance of a sterile environment is essential. For the production of mushroom spawn at home, the main methods for sterilizing tools and growth media are disinfectant, boiling and flames.

Tools such as knives and tweezers can be sterilized by placing the tips or blades into the flame of a bunsen burner or a similar clean heat source. Jars and growth media can be boiled for at least an hour to sterilize them. The work environment can be cleaned with a 70 percent solution of ethanol to remove contaminants.

When everything is clean and sterile, the mother culture prepared from spores and the mycelium in the millet grains will exhibit strong, healthy growth for a high-quality mushroom spawn.
